Specificity

Clone 1J22 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Calpain -9. It targets an epitope within 21 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal half of human Calpain-9.

Application

Quality Control Testing

Evaluated by Western Blotting in HT-29 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Calpain-9 in HT-29 cell lysate.

Tested applications

Affinity Binding Assay: A representative lot of this antibody bound Calpain-9 with a KD of 1 x 10-12 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Calpain-9 in HT-29 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Calpain-9 in human stomach tissue sections.

Anti-Calpain-9, clone 1J22 ZooMAb®, Cat. No. ZRB1383, is a recombinant Rabbit monoclonal antibody that specifically targets Calpain-9 and is tested in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Calpain-9 (UniProt: O14815; also known as Digestive tract-specific calpain, New calpain 4, nCL-4, Protein CG36) is encoded by the CAPN9 (also known as NCL4) gene (Gene ID: 10753) in human. The mammalian calpains include both ubiquitous and tissue-specific members. Two major ubiquitous calpains, m-calpain and μ-calpain, and two gastrointestinal-tract (GI) specific calpains, calpain-8 and calpain-9 are found in stomach and intestine. Calpains-9 has a typical domain structure that is similar to calpains 1 and 2. It has a regulatory N-terminal domain (domain I), a protease domain (II, consisting of sub-domains IIa and IIb), a C2-like Ca2+/phospholipid-binding domain (III), and a penta-EF-hand (PEF) domain (IV). Its catalytic region is localized in amino acids 43-337. Calpain-9 expression is down-regulated in gastric cancer tissue and in gastric cell lines of differentiated and poorly differentiated types. It is considered as a tumor suppressor and its low expression has been positively associated with late T stage (tumor size), lymph node metastasis, and advanced TNM stage. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
